生理機能が不明な受容体

オーファン とは 孤児 のことで、親 情報の化学的実体 というべき機能性の 生体内リガンド ホルモンなど が同定されていないことから、このように呼ばれま す。 当研究室では主に 受容体 Gα融合タンパク質を用いたリガンド検索 とい う方法を用いています。 Gタンパク質共役受容体 GPCR は三量体Gタンパク質と共役して働き、現在使用されている臨床薬の約５割以上がGPCRをターゲットにしている といわれている。 多くの方法が開発された中でもっとも成果をあげ、広く用いられている方法はアゴニスト GPCR Gタンパク質 特にGq系 PLC IP3 細胞 内カルシウム濃度上昇という一連の変化で起こる変化を、細胞に導入したFura-2やFluo-3などの蛍光の変化で観測する方法である。 S]-GTPγS結合活性を測定する系を利用することとした.融合タンパク質を含む膜画分にアゴニストを加える と受容体につづいてGタンパク質αサブユニット部分が活性化されるが、GαにはGTPase活性があるので活性化に際してGαサブユニットからの GDPの解離が促進されGTPが結合する。

ヒトゲノムプロジェクトによって明らかにされた遺伝子配列からGPCRをコードすると思われる未知の 遺伝子が多数見つかってきており、これらはその受容体が結合するリガンド 身体の中に存在するホルモンなど が不明であることから オーファン GPCR 孤児受容体 と呼ばれます。

I have travelled tons this summer and having been working on a big project (more on that later), hence the lag with the blog. I was in NYC this summer and spent an afternoon at the MOMA. I had my camera with me and was intrigued by the amount of people taking photos of the art work. As the camera screens were surrounding me, I decided to document my experience in a different way.
